Bonamia ostreae, parasite of the European flat oyster, Ostrea edulis, does not experimentally infect the Japanese oyster, Crassostrea gigas ArchiMer

The protozoan Bonamia ostreae (Ascetospora) is the etiological agent of bonamiosis, or hemocyte disease of the European fiat oyster, Ostrea edulis (Pichot et al., 1980). Since this infection was first reported from the coast of Brittany (Comps et al., 1980), the parasite has spread rapidly along the French Atlantic coasts (Grizel & Tigé, 1982), and in the European countries including the UK, Holland, lreland and Spain (Bannister & Key, 1982; Van Banning, 1982; Polanco et al., 1984). Under the EEC Council Directive 91/67, this parasitic protozoan is included in list II of Annex A. Serious pathogen agents for molluscs that should be obligatorily declared are placed in this list

The parasite, Bonamia ostreae, was diagnosed in several South Brittany flat oyster's samples, from gills tissues's prints, stained in May-Grunwald-Giemsa and from corresponding digestive glands sections. The comparaison of results shows quantitative differences relating to the infested oyster's rate, and qualitative differences relating to the early stages of infestation. Gill's print technic seems more intersting for the only Bonamia ostreae diagnosis.

Can survival of European flat oysters following experimental infection with Bonamia ostreae be predicted using QTLs? ArchiMer

The present study identifies quantitative trait loci (QTLs) in response to an experimental infection with the parasite responsible for bonamiosis, Bonamia ostreae, in two segregating families of the European flat oyster, Ostrea edulis. We first constructed a genetic-linkage map for each studied family and improved the existing genetic-linkage map for the European flat oyster with a set of SNP markers. Bonamia ostreae is an intracellular protistan parasite affecting flat oysters Ostrea edulis. It can be detected in juveniles but mortalities mainly affect oysters which are more than 2 years old. The parasite is usually observed inside haemocytes and sometimes free, notably in gill epithelia suggesting a parasite release through this organ. However, the infective form and ways of entry and release remain undetermined. Flat oysters incubate their larvae in their pallial cavity for 8–10 days before releasing them into the water column. Les méthodes de classification sont largement utilisées en vision par ordinateur et reconnaissance des formes. La classification floue a d'ailleurs montré de bonnes performances dans ces domaines, non seulement dans la reconnaissance de classes en volume, mais aussi pour des formes fines, des droites, des courbes, et également des surfaces. La morphologie et l'équipement enzymatique des hémocytes de deux espèces d'huîtres, Ostea edulis et Crassostrea gigas ont été étudiés. L'analyse des fonctions des hémocytes dans les mécanismes immunitaires a permis de mettre en évidence leur rôle de macrophage, mais n'a pas fait apparaître de différence interspécifique dans les phénomènes des cytocidie oxygène dépendants, susceptible d'expliquer la permissivité d'Ostrea edulis pour Bonamia ostreae et la résistance de Crassostrea gigas. Additional genetic markers are required to improve population genetics study and linkage map development for selection for B. ostrea-resistance in this species. Here, we characterized 27 novel microsatellite loci for O. edulis. Number of alleles per locus ranged from 6 to 25 and observed heterozygosity between 0.375 and 1. Null alleles were suggested at a few loci but most loci were in Hardy-Weinberg agreement enabling their reliable use in further population and mapping genetics approaches.

Combining Two-Stage Testing and Interval Mapping Strategies to Detect QTL for Resistance to Bonamiosis in the European Flat Oyster Ostrea edulis ArchiMer

We have identified quantitative trait loci (QTL) in the flat oyster (Ostrea edulis) for resistance to Bonamia ostreae, a parasite responsible for the dramatic reduction in the aquaculture of this species. An F-2 family from a cross between a wild oyster and an individual from a family selected for resistance to bonamiosis was cultured with wild oysters injected with the parasite, leading to 20% cumulative mortality. As of 1985, Ifremer set up a selection programme for resistance to the protozoan Bonamia ostreae in European flat oysters (Ostrea edulis). Two selection lines, L85 and L89, from the third and second generation, respectively, were produced with 17 and 21 different families. The experimental design also included a mixed line (L85 x L89) of seven families and a control line of five families from Quiberon broodstock maintained without selection pressure.

Farming of the flat oyster Ostrea edulis in Europe is severely constrained by the protozoan Bonamia ostreae. The introduction of the resistant species Crassostrea gigas has been a relief for the farmers, while the pilot programmes to select O. edulis strains resistant to bonamiosis performed in various countries can be seen as a promising strategy to minimise the effects of bonamiosis. Compléments d'observations sur le cycle du parasite, Marteilia refringens (Grizel et coll. 1974) ArchiMer

Les premiers stades connus du parasite ont été mis en évidence en Bretagne pendant les mois de juillet et d'août (GRIZEL et TIGE, 1977). Suite à ces observations, un essai a été réalisé en 1978 pour savoir si l'infestation avait bien lieu à ces dates ou si elle se faisait antérieurement sous une forme non décelée actuellement et subissant des transformations profondes dans les tissus de l'huître. Les résultats montrent que l'infestations se fait effectivement pendant l'été, à partir du mois de juillet jusqu'à la fin du mois d'août. Elle fut dans l'ensemble faible; les lots les plus infestés sont ceux qui ont séjourné le plus longtemps en zone contaminée.

La discussion générale organisée à la fin de la deuxième journée a fait ressortir les points suivants: 1- Les participants se sont déclarés satisfaits de la tenue de la réunion, tant dans son déroulement, que dans les échanges d'informations auxquels elle a donné lieu. La quantité et la qualité des résultats présentés a permis de mettre en évidence la puissance d'analyse que représente le travail en réseau. 2-L'importance de la valorisation des résultats a été soulignée à plusieurs reprises. Le programme de sélection de l'huître plate a été initié en 1985 par des pathologistes de la Tremblade. La sélection ne portait que sur le seul critère de tolérance des huîtres plates au parasite Bonamia ostreae et correspondait plus à un outil pour étudier divers aspects de la parasitose qu'un un programme d'amélioration génétique visant la relance de la culture de l'huître plate. Dans le cadre du réseau REGEMO, ce programme a ensuite été repris par les généticiens de l'Ifremer : 1) rappel historique et définition d'un vocabulaire commun à l'ensemble des membres du réseau REGEMO. 2) Inventaire et bilan des opération menées ou en cours ( sur la dernière génération de sélection produite ne 1995, sur le matériel issu de cette génération et produit en 1997....

Compte-rendu des résultats d'analyses effectuées en histologie - Année 1995 ArchiMer

Dans le cadre du Réseau zoosanitaires CEE (huître plate) et suivi d'autres espèces, 890 individus provenant des zones 5, 6 et 7 ont été examinés sur l'ensemble de l'année 1995 par différentes techniques : 472 analyses sous coupes histologiques, 328 analyses sur appositions d'organes, 90 analyses sur milieu au thioglycolate.
